Genetics Shape Mood Disorders: Insights from Recent GWAS Reveal Distinctions in Bipolar Types and Their Links to Depression

Jonathan Coleman, PhD, a statistical geneticist and postdoctoral fellow, works in Gerome Breen’s lab at King’s College London. “The clearest findings are a genetic distinction between type 1 bipolar and type 2 bipolar, and the greater similarity of type 2 bipolar to major depressive disorder,” Coleman stated.

Manic-depressive disorder was the term once used to describe both forms of bipolar illness. A manic state features heightened behavior, euphoric or irritable mood, decreased sleep needs, impulsive actions, impaired judgment, racing thoughts, disordered thinking, and often vivid hallucinations or erroneous beliefs. Type 1 bipolar disorder involves both mania and depression, while Type 2 bipolar disorder primarily involves depression accompanied by moderate symptoms resembling mania, known as hypomania.

Genetic Insights from Massive Datasets Reveal Disparities and Unexpected Connections in Mood Disorder Risks

The conclusions were drawn from a combination of numerous very large datasets. Coleman, Breen, and their co-authors examined both common and unique molecular genetic connections in their meta-analysis by combining genome-wide association studies from three sizable datasets of individuals with major depression and bipolar illness. The vast international Psychiatric Genomics Consortium provided the majority of the data. Researchers gathered additional information from 23andMe, an online genetic service platform, and the UK Biobank, a significant health resource founded by the Wellcome Trust.

The results of genome-wide association studies (GWAS) highlight notable racial and ethnic disparities. The study’s findings on genetics and mood disorders apply only to Europeans, with potential differences in other populations.

The authors found that genetic risk for these disorders predicts other features. Higher educational attainment links to bipolar disorder risk, while lower attainment links to major depressive disorder risk.

Scientists mapped hereditary susceptibility to these illnesses to specific brain cells in mice using gene expression pattern analysis. Depression and bipolar disorder link to serotonin neurons, but GABA and glutamate neurons specifically involve bipolar disorder.

John Krystal, MD, notes that mood disorders are highly varied, with clinically difficult-to-define boundaries between their types. Consequently, this highlights mood disorders’ complexity and the challenges in diagnosis and treatment, as their distinctions are often unclear.

Krystal notes the study suggests genetic risk and brain function link various mood disorder forms. Consequently, this implies underlying genetic and neurological factors connect mood disorders, suggesting a common thread in their development and manifestation.

Krystal adds that distinctions could reveal depression subtypes with significant implications for treatment approaches and effectiveness. Moreover, understanding these differences could enable targeted treatments for specific depression subtypes, improving patient outcomes and advancing treatment methods.

The research aims to develop tools to predict if an initial depressive episode will persist or evolve into bipolar disorder. Coleman stated genetic data won’t replace clinical insight but could be a valuable addition to clinical models.
